Determining the intrinsically disordered regions of proteins is crucial for elucidating their biological functions and the mechanisms of related diseases. However, current single-sequence-based methods have low accuracy, while evolution-profile-based methods are computationally intensive. Therefore, we propose a highly accurate protein disorder predictor, ProtSSSD, which utilizes the protein language model along with a LoRA fine-tuning strategy and incorporates a multitask learning architecture. The results show that ProtSSSD outperforms all single-sequence-based methods and achieves better performance than two other language model-based techniques in the second Critical Assessment of protein Intrinsic Disorder prediction experiment. Moreover, ProtSSSD demonstrates comparable or even superior performance to the state-of-the-art profile-based method SPOT-Disorder2.
